Multifocus microscopy with optical sectioning and high axial resolution

چکیده
Multifocus microscopy enables recording of entire volumes in a single camera exposure. In dense samples, multifocus is severely hampered by background haze. Here, we introduce scalable method that incorporates optical sectioning and offers improved axial resolution capabilities. our method, dithered oblique light-sheet scans the sample volume during exposure, while fluorescence from each illuminated plane mapped onto line on with element. A synchronized rolling shutter readout realizes sectioning. We describe technique theoretically verify its improvement demonstrate prototype system beam splitter cascade record monolayers endothelial cells at 35 per second. furthermore image uncleared engineered human heart tissue visualize distribution mitochondria high resolution. Our manages to capture sub-diffraction sized mitochondria-derived vesicles up 30 µm deep into tissue.

Axial gain resolution in optical sectioning fluorescence microscopy by shaded-ring filters.
We present a new family of pupil masks to control the axial component of the intensity distribution in the focal region of tightly focused light fields. The filters, which consist of a circular clear pupil with a single shaded ring, allow to control the width of the central lobe of the axial spot together with the residual sidelobes energy. The filters can be applied to improve the optical sect...
متن کاملDMD-based LED-illumination Super-resolution and optical sectioning microscopy
Super-resolution three-dimensional (3D) optical microscopy has incomparable advantages over other high-resolution microscopic technologies, such as electron microscopy and atomic force microscopy, in the study of biological molecules, pathways and events in live cells and tissues. We present a novel approach of structured illumination microscopy (SIM) by using a digital micromirror device (DMD)...
متن کاملThe OPFOS Microscopy Family: High-Resolution Optical Sectioning of Biomedical Specimens
We report on the recently emerging (laser) light-sheet-based fluorescence microscopy field (LSFM). The techniques used in this field allow to study and visualize biomedical objects nondestructively in high resolution through virtual optical sectioning with sheets of laser light. Fluorescence originating in the cross-section of the sheet and sample is recorded orthogonally with a camera. In this...
متن کاملHigh-resolution optical diffraction microscopy.
In an optical diffraction microscopy experiment, one measures the phase and amplitude of the field diffracted by the sample and uses an inversion algorithm to reconstruct its map of permittivity. We show that with an iterative procedure accounting for multiple scattering, it is possible to visualize details smaller than lambda/4 with relatively few illumination and observation angles. The roles...
متن کاملWhole-field five-dimensional fluorescence microscopy combining lifetime and spectral resolution with optical sectioning.
We report a novel whole-field three-dimensional fluorescence lifetime imaging microscope that incoporates multispectral imaging to provide five-dimensional (5-D) fluorescence microscopy. This instrument, which can acquire a 5-D data set in less than a minute, is based on potentially compact and inexpensive diode-pumped solid-state laser technology. We demonstrate that spectral discrimination as...
